Human Senile Cataractous Lens Protease
نویسنده
چکیده
A proteolytic enzyme was isolated from human senile cataractous lens by anion-exchange and gel-filtration chromatography. Sedimentation and zoneelectrophoretic experiments indicated a high degree of homogeneity for the enzyme. A molecular weight of 27000 was calculated from measurements of sedimentation velocity and diffusion coefficient. Chelating agents decreased activity which could be restored by addition of certain bivalent metal ions. Diisopropyl phosphorofluoridate and phenylmethanesulphonyl fluoride inhibit the proteolytic activities. Optimum rates of hydrolysis were observed at pH5.2.

Lens superoxide dismutase and catalase activities in diabetic cataract.
OBJECTIVE Biochemical evidence suggests that the oxidative damage of the lens proteins is involved in the genesis of senile cataract and the development of diabetes-related pathologic changes such as the formation of cataracts. In particular, lens proteins are subject to extensive oxidative modification. Oxidative damage either decreases the antioxidant capacity or decreased antioxidant capacit...
متن کاملIncrease in lens gangliosides due to aging and cataract progression in human senile cataract.
Gangliosides were isolated from human senile cataractous lenses by solvent extraction, DEAE-Sephadex column chromatography, and thin-layer chromatography. The content and composition of gangliosides were examined in individual lens tissues. Three predominant gangliosides, GM3, GM1, and GD1a, were tentatively identified in comparison with authentic brain gangliosides, and several unidentified ga...
متن کاملHuman lens epithelium in normal and cataractous lenses.
Epithelial cell appearance and cell size were studied in intact, unfixed, and unstained human lenses. Ten normal and 24 cryoextracted senile or presenile cataractous lenses, all with subcapsular cataract, were examined. The cell size of the transparent normal lenses showed little variation. However, in the cataractous lenses, a wide range was found in mean cell size of different lenses and vari...
متن کاملRole of lipid peroxidation in the pathogenesis of myopic and senile cataract.
AIMS/BACKGROUND Increased production of free radicals, consumption of antioxidant, and oxidation of unsaturated lipids have been observed recently in cataractous lenses and active participation of the retina in human cataractogenesis has been proposed. To verify this hypothesis, the total (GSH) and oxidised (GSSG) glutathione concentrations were assayed in the lens and the malondialdehyde (MDA)...
متن کاملStudies on protein and taurine in normal, senile and diabetic cataractous human lenses.
An attempt has been made to explore the possible role of Taurine in cataractogenesis. Normal lenses were obtained from eye bank donors and cataractous lenses from patients who had undergone surgery for cataract extraction. Lenses were weighed and homogenised. Extraction, isolation and estimation of protein and taurine were carried out. It has been found that the lens wet weight increased progre...
